This first, very modest hospital was created two years after the founding of the city under the name of Hospital San Sebastián, intended to provide for the first emergencies. It was replaced by a larger and more functional building in 1585, which was taken over by the Order of St. John of God in 1620, with four nuns arriving from Lima. The hospital was a large house of lime and stone, with a church, cloister, outbuildings and a military hospital.
